چکیده

Transcription factor GATA-1 is essential for the development of the erythroid lineage. To ascertain whether strict control of GATA-1 expression level is necessary for achieving proper erythropoiesis, we established transgenic mouse lines expressing green fluorescent protein (GFP) under the control of the GATA-1 gene hematopoietic regulatory domain. We examined the GATA-1 expression level by exploiting the transgenic mice and found two GFP-positive hematopoietic progenitor fractions in the bone marrow. One is the GFP fraction containing mainly CFU-E and proerythroblasts, which coexpress transferrin receptor, while the other is the GFP/transferrin receptor-negative fraction containing BFU-E. Since the intensity of green fluorescence correlates well with the expression level of GATA-1, these results indicate that GATA-1 is highly expressed in CFU-E, but low in BFU-E, suggesting that the incremental expression of GATA-1 is required for the formation of erythroid progenitors. We also examined GFP-positive fractions in the transgenic mouse spleen and fetal liver, and identified fractions containing BFU-E and CFU-E, respectively. This study also presents an efficient method for enriching the CFU-E and BFU-E from mouse hematopoietic tissues. (E-mail, [email protected]) 2 only. Changing Pattern of Histone H3 Methylation following treatment of erythroid progenitors derived from cord blood CD133+ cells with sodium butyrate and thalidomide

Background: Human β-like globin genes regulaon during development from embyonic to adult stage results in generaon of different types of hemoglobin with different funcons. As β-thalassemia and sickle cell disease are disorders of β-globin chain, epigenec drugs such as thalidomide and sodium butyrate which can induce γ-globin gene are considered as a novel therapeuc approach. Drugs effec...

Identification and characterization of three classes of erythroid progenitors in human fetal liver.

Erythroid progenitors from 1 8 fetal livers (7-1 5-wk gestational period) and 1 3 normal adult marrows were grown in methylcellubose cultures containing erythropoietin (Ep) and burst-enhancing factor(s) (BEF). Additional experiments were carried out on 1 fetal, 1 5 neonatal. and 26 adult blood specimens. Three classes of progenitors (primitive or mature erythroid burst-forming unit(s). P-BFU-E....

Adult Hippocampal Neurogenesis and Memory

Adult neurogenesis, a concept emergent in the late 1990s, is the generation of new neurons in the adult brain. This process occurs thank to cells who have this proliferative feature, named as Neural Stem Cells (NSCs). Neural Stem Cells (NSCs) are primary progenitors who can generate the two neural types (neurons and glia). Classically it was assumed that NSCs are only present in the embryo, but...

Isolation and Characterization of Human Induced Pluripotent Stem Cells-Derived Mesenchymal Progenitors

Purpose: Isolating human induced pluripotent stem cells (hiPS)-derived mesenchymal progenitors as a new source of mesenchymal cells which can differentiate into different lineages like adipose and bone. Materials and Methods: After 7 days of hiPS1 culture on matrigle coated dishes, spindle like cells around colonies were removed by cell scraper. These cells that had mesenchymal like morphology ...
